L'Autore attacca ogni forma di pregiudizio, con particolare insistenza su quelli legati alle credenze religiose e alla sfera del potere politico, considerandoli insieme all'ignoranza e all'errore la fonte dei mali dell'uomo, mentre "la vérité en est le remède". Il filosofo illuminista non esitava a riconoscere l'interesse del clero e della tirannia a mantenere l'umanità nell'abisso dell'ignoranza attraverso strumenti come il peso oppressivo della religione, l'iniquità delle leggi e il controllo arbitrario dei privilegi della corte. L'opera suscitò la reazione ostile di Federico il Grande, che ne rifiutò le tesi nel suo Examen de l'Essai sur les préjugés: contro le posizioni del sovrano prussiano si schierò a sua volta Denis Diderot nella Lettre sur l'Examen de l'Essai sur les préjugés ou Pages contre un tyran.
